Although herbal medicines rely on remedies of natural origin and have fewer side effects than allopathic medicines, the use of herbal remedies is not always safe. This is due to the lack of information provided about bioactive constituents, extraction methodology, and purity of these natural products. Eurycoma longifolia (Tongkat ali), is a popular traditional herbal medicine native to Malaysia, Indonesia, Vietnam and also Cambodia. The root extracts of this plant is heavily used mainly for its aphrodisiac properties. However the this extracts have also been reported to be effective as antibiotics, combating various illnesses, constipation, fever, and generally as health supplement. In our study, we highlighted the effectiveness of Tongkat ali fractions as anti-parasitic agent against various subtypes of Blastocystis sp. This fraction was then furthered for the analysis of potential active principles using liquid chromatography mass spectrometry (LCMS/MS). This analysis was conducted using AB Sciex 3200QTrap LC-MS/MS with Perkin Elmer FX 15 uHPLC system, equipped with an Agilent Zorbax C18 column (150mm x 4.6mm x 5µm). A total of 10 compounds were identified from the water fraction consisting of quassinoids, β-carboline alkaloids and canthin-6-one alkaloid. Some of these compounds, namely, eurycomanone and Laurycolactone B have previously been reported to exhibit anti-protozoal properties which deduces the effectiveness of Tongkat ali as anti-protozoal agent against Blastocystis sp.
